原理介紹

無銅點(diǎn)擊化學(xué)反應(yīng),即菌株促進(jìn)的疊氮炔環(huán)加成(SPAAC)是一種生物正交反應(yīng),利用一對試劑,即環(huán)辛基和疊氮化合物,它們彼此有效地反應(yīng),同時(shí)對羥基、氨基和硫醇等天然官能團(tuán)保持惰性。SPAAC能夠通過形成穩(wěn)定的三唑,在水和其他復(fù)雜的化學(xué)環(huán)境中標(biāo)記各種各樣的生物分子,而無需任何輔助試劑。在眾多已知的環(huán)辛基化合物中,二苯并環(huán)辛基化合物(DBCO)是一類具有相當(dāng)快的動力學(xué)和良好的水緩沖穩(wěn)定性的試劑。在生理溫度和pH范圍內(nèi),DBCO基團(tuán)不會與許多生物分子中天然存在的巰基、氨基或羥基發(fā)生反應(yīng)。DBCO-PEG4-琥珀酰亞胺酯是一種常用的通過氨基修飾將DBCO基團(tuán)引入生物分子的試劑。靶點(diǎn)科技分銷的ClickChemistryTools的DBCO-PEG4-NHS Ester,是高效率的DBCO 琥珀酰亞胺酯。
